چکیده

In this article we present data that compares RNasin® Ribonuclease Inhibitor to SUPERase•InTM, an RNase inhibitor recently introduced by Ambion. The two RNase inhibitors are compared for their ability to protect RNA from degradation under a variety of different reaction conditions. In addition, we tested both inhibitors for their ability to protect RNA during in vitro translation and transcription.

RNasin® Ribonuclease Inhibitor Part I: Characterization of the Protein, PN077

Ribonucleases (RNases) are ubiquitous enzymes that can detrimentally affect RNA isolation in the laboratory. Many applications such as RT-PCR and in vitro transcription and translation may be affected as well. RNase can renature after autoclaving and is difficult to inactivate. Many RNase inhibitors are available commercially. Promega’s RNasin® Ribonuclease Inhibitor, one of the first introduce...

Characterization of Ribonuclease Activity of Three S-Allele-Associated Proteins of Petunia inflata.

Three S-allele-associated proteins (S-proteins) of Petunia inflata, a species with gametophytic self-incompatibility, were previously found to share sequence similarity with two fungal ribonucleases, RNase T(2) and RNase Rh. In this study, the S-proteins from P. inflata plants of S(1)S(2) and S(2)S(3) genotypes were purified to homogeneity by gel filtration and cation-exchange chromatography, a...

Characterization of the ribonuclease activity on the skin surface

The rapid degradation of ribonucleic acids (RNA) by ubiquitous ribonucleases limits the efficacy of new therapies based on RNA molecules. Therefore, our aim was to characterize the natural ribonuclease activities on the skin and in blood plasma i.e. at sites where many drugs in development are applied. On the skin surfaces of Homo sapiens and Mus musculus we observed dominant pyrimidine-specifi...

Methylation sensitivity of restriction enzymes interacting with GATC sites.

924 BioTechniques Vol. 34, No. 5 (2003) inates the residual ribonuclease activity, although the effect of this treatment on the subsequent experiments should be checked. An inhibitor of RNase, such as Rnasin, can also be added to further enzymatic reactions. This study has described a degrading activity in RNA preparations from ovine, bovine, and porcine brain and ovine liver. Further experimen...

بررسی فعالیت مهارکننده ریبونوکلئاز جفتی بر روی آنزیم ریبونوکلئاز

Ribonuclease inhibitor is an acidic protein, soluble in cytoplasm. It is abundantly present in human placenta. It could be extracted from this organ to the extent of 1mg/placenta using chemical ion exchange and affinity techniques. Molecular weight of this protein is estimated by electophoresis or SDS-PAG electrophoresis using standards of known proteins. Molecular weight has been found to be 4...
